Abstract

The in vitro regeneration ability of Patchouli shoot tip and leaf explants was studied and a simple defined medium has been standardized for mass multiplication of plants. The shoot tip cultured on MS medium was directly regenerated in to shoots and the shoots when supplemented with 2.0 mg/l BAP and 0.1 mg/l NAA exhibited higher percentage of proliferation and registered maximum shoot weight (15.90 g). Indirectly the leaf explants was induced to callusing and further regenerated in to shoot formation on the same medium supplemented with 2.0 mg/l 2, 4-D and 0.1 mg/l BAP that had produced maximum callus weighing 55.30 g. The regenerated shoots were readily rooted on half strength MS medium and IAA has had greater influence on number and length of roots. The rooted plantlets hardened on medium having soil-rite, and sand and soil-rite in the ratio of 1:1 were established with a cent per cent survival rate.
